Dedicated laboratory space is located within the Science and Engineering I building at UC Merced. The lab is well equipped with microscopy facilities in two dedicated microscope rooms, plus wet chemistry space. Equipment includes a custom-built optical trapping system. The optical trap is integrated with a force-sensing module, a three-dimensional nano-positioning stage, differential interference contrast microscopy, and epi-fluorescence microscopy. The system is housed in a Nikon Ti- U inverted microscope, and vibration-isolated with a Newport SmartTable. The laboratory also houses a Nikon TIRF. The motorized TIRF attachment couples a 488-nm excitation line (120 mW, Coherent) to a Nikon Ti-E inverted microscope. The system is integrated with the Nikon Perfect Focus System, epi- fluorescence capability, coupled to an Andor EMCCD via a motorized filter wheel, and vibration-isolated with a Newport Vision IsoStation. Three data-acquisition PCs (i7 and i5 processors) are available. Each PC is equipped with solid-state hard drives for sustained data recording at 70 Hz. Data are achieved using custom-developed programs in LabVIEW (available on all PCs) or in Nikon NIS-Elements (one PC). Four data-analysis PCs (i7 and i5 processors) are available. Each PC is equipped with particle-tracking and velocity-parsing routines in Virtualdub and in MATLAB. Other major equipment includes freezers (- 80 °C, -20 °C), refrigerators (4-8 °C), centrifuges, an ultra-centrifuge, an oven, a laminar flow hood, a sonicator, and precision scales.
